Equipements plateforme Biochimie


The Platform has high performance equipment grouped into four categories: production and purification, centrifuges, analytical techniques and biomolecular interactions.

Production and purification


An incubateur Innova 42 (Eppendorf) with control of the temperature of 4 to 80ºC.

Two grinders of cells :
POLYTRON with a probe of up to 30000 rpm.
FastPrep24 (MP Biomedicals). Grinding the samples in a few seconds by using beads of different sizes and composition according to the sample. A wide range of samples can be used (tissues, bacteria, eukaryotic cells, bone, wood ...).

A Micropulser electroporation (Biorad). Introduction of DNA into cells by application of an electric field

The RTS Proteomaster (Roche) which allows to obtain small amounts of protein without production in organism, only with kits from Roche.

A FPLC system (Akta Purifier, GE Healthcare), refrigerated at 4ºC which allows the realization of high performance liquid chromatography for fast and precise separation of proteins.

Centrifuges


Several types of centrifuge / ultra-centrifuges (Beckman Coulter) are available under reservation

Analysis techniques


The platform includes the equipment necessary for the realization of single and two-dimensional electrophoresis gels and Western blots (Biorad).


Le Trans-Blot Turbo Transfert System (Biorad) allows a transfer of the proteins on the membrane in 5 to 30 minutes depending on the type of gel used.

The Platform has several analysis systems


Two detection systems for the analysis of acrylamide gel and blots by colorimetry and chemi-luminescence. G:Box iChem (Syngene) et ChemiDoc MP (Biorad).

Plate reader Model 680 (Biorad) for measurement between 400 and 750 nm. Ideal for determinations of protein concentration by colorimetry.

Plate reader POLARstar Omega (BMG Labtech), multidetection reader equipped with an injection system for a UV/Vis but also fluorescence (FRET, polarization), luminescence, and LVIS plate for quantifying DNA, RNA and protein by absorbance.

Plate reader MAGPIX (Biorad), to multiplex ELISA experiment that works with magnetic beads and which can identify and quantify 50 target molecules per well on 96 well plate. Many kits are available but it is also possible for you to make them.

A protein quantification apparatus based on a measurement in the infrared: the Direct Detect Spectrometer (Merck Millipore). Only 2µL are necessary.

A Zetasizer uV (Malvern) allows the determination of the size (0.3-2000 nm) of purified proteins or particles.

THe Ingenuity Software (QIAGEN).
This is a manually annotated bank that allows to quickly analyze and interpret transcriptomic and proteomic data. It is also a very effective bibliographic tool.

The Nanodrop-one (Ozyme) quantify and qualify DNA, RNA, and protein samples with only 1-2 µL.

qNano (IZON) directly measures concentration and particle diameter (50nm to 10um).

Biomolecular interactions


The isothermal titration calorimetry apparatus (ITC) of the platform allows the analysis of biomolecular interactions by a biophysical process measuring heat exchange.


A ITC200 (Malvern) allows the study of molecular interactions by isothermal titration calorimetry. This device allows to determine the affinity constant, stoichiometry and thermodynamic parameters of the reaction from purified partners (proteins, peptides, sugars, lipids, metal ions, DNA, RNA, ...)

This system measures the heat released or absorbed during the interaction between the two partners. The reaction may be exothermic (releasing heat) or endothermic (heat absorbing). One of the molecules is injected at regular intervals in the measuring cell which contains its partner until saturation of the binding site.
The curve allows to obtain the affinity constant (Kd), the stoichiometry (N) of the reaction and the thermodynamic parameters (ΔH and ΔS, enthalpie and entropie).




Water purification systems


Two water purification systems (Millipore) produce distilled water and MilliQ water on the platform.
